Xin chào,

• Mình sử dụng máy hplc của shimadzu chạy thấy đường nền bị nhiễu, mình đã rửa máy bằng nước, rồi bằng 2-propanol [không qua cột] vẫn không hết, cột đó đem qua máy khác vẫn bình thường. Mình phải làm sao.
• mình có cây cột hplc chạy nhiều rồi nên peak rất xấu, mình có thể xử lý được không [cột mình chạy trong dung dịch đệm với acetonirile hoặc đệm với methanol] Ai biết chỉ mình với

Hi, Cái cột đầu tiên Có thể bộ phận máy của bạn bị dơ, bạn vệ sinh máy thử xem. Cái cột thứ hai Ban rửa cột lại cho sạch, rồi lật ngược có lại để chạy (đảo đầu). Thân!

Hi! đường nền nhiễu bạn thử kiểm tra xem flow cell có bị dơ không ( rút ra và nhìn xuyên qua hai mặt kính) nếu OK thì bạn thử kiểm tra giờ đèn đã sử dụng, và kiểm tra cường độ đèn xem có đạt không nhé!!! thân

Trước tiên cho mình hỏi hệ thống của bạn mua lâu chưa? Bơm của bạn hoạt động tốt không? Độ giao động của bơm như thế nào? Nhiễu nền giữa các lần chạy khác nhau trên cùng một mẫu với một chế độ chạy có như nhau không? Độ nhạy của peak cần xác định so với ban đầu có giảm không?

Như bạn nói thì cột của ban đem qua máy khác chạy thì tốt, điều đó chứng tỏ cột của bạn tốt.

Vấn đề còn lại chỉ có thể do pump hay detector thoi. Detector bạn vài loại nào, nếu bạn xài UV-Vis, PDA, bạn có thể clean flow cell để xem nó còn bị nhiễu không theo như cách dưới đây thử xem sau:

[b][i]Occasionally, eluted compounds may deposit on the cell walls, thus increasing the detector noise level.

1. Flush the flow cell with HPLC-grade methanol and observe the baseline.
2. If this procedure does not solve the problem, flush the cell with 0.1M nitric acid. Warning: To avoid damage to the skin and eyes, wear appropriate protective clothing and goggles when using nitric acid. 3.Flush the flow cell with HPLC-grade water until the solvent leaving the cell is neutral (pH 7).
3. If cleaning the flow cell does not eliminate the problem , install a new flow cell.[/i][/b]

PS: Ah, cái cột kia của bạn nên rửa lại nhiều lần thử xem thế nào, còn đảo cột là cách cuối cùng, khi không còn làm gì được nữa thôi, đảo cột nếu chạy tốt thì sẽ được một thời gian nữa, nếu không thì bỏ cột luôn.

• Bạn nên rửa lại hệ thống bằng MeOH:H2O 50/50 vài lần đi. Sau đó, kiểm tra flow cell theo gợi ý của complexchemistry và quy trình của <st1:place w:st=“on”><st1:city w:st=“on”>Tokyo</st1:city></st1:place> để kết luận. Ngoài ra bạn kiểm tra đèn D2 xem, của Shimadzu thì life time của đèn D2 > 500 h là gần như không dùng được rùi. Nếu loại bỏ 3 yếu tố trên thì bạn mới cần gọi kỹ sư đến để kiểm tra hệ thống và hệ quang trong Det.
• Nếu cột chạy nhiều rồi thì bạn rửa đi bằng MeOH xem sao (khi rửa thì tháo đầu nối và Det ra nhé lấy cái cốc mà đựng dm rửa. Nếu không được thì bạn tháo đầu của cây cột đó ra, đổ bớt đi 1 ít chất nhồi (silica gel, aluminia oxit,…) là dùng lại được tương đối đó.

trước hết bạn phải biết loại cột bạn đang chạy là cột gì. tương tác như thế nào để có dung môi rửa cột cho phù hợp. ví dụ với loại cột C18 dung môi thường dùng rửa cột là MeOH nguyên chất. nếu không được có thể dùng đến acetonitrin.
